Barsana vs Vrindavan: the short answer
Barsana and Vrindavan are two beloved but different towns of Braj and they belong together in one pilgrimage. Vrindavan is the main town of Braj, the town of Krishna's childhood, on the sacred Yamuna, filled with thousands of temples and joyful bhakti; it is the hub where most pilgrims stay. Barsana, about 45 to 50 km to the west, is the hill town of Radha Rani, Krishna's beloved, crowned by the great Radha Rani temple and famous for the Lathmar Holi; it is usually seen as a day trip. So Barsana is not the same as Vrindavan; it is Radha's own town, visited from Vrindavan. To see both and the whole of Braj, a mathura vrindavan tour with Experience My India can plan it.
From the ground (Gurudutt): People sometimes think Barsana and Vrindavan are the same, but they are two towns, about an hour and a half apart. Vrindavan is Kanhaiya's town, the big one, where you stay, with all the great temples; Barsana is our Kishori Ji's hill town, Radha's own, where you go for a day, above all for Radhashtami and the Lathmar Holi. Stay in Vrindavan and let me take you to Barsana; you need both to know Braj. Radhe Radhe.
What Vrindavan is
Vrindavan is the main town of Braj, the heart of Krishna devotion:
Krishna's town and the hub. Vrindavan, on the sacred Yamuna, is the town of Krishna's childhood and the main hub of Braj, where most pilgrims stay; from here they visit the wider Braj, including Barsana, Nandgaon and Govardhan.
A land of temples. Vrindavan holds thousands of temples, from the beloved Banke Bihari and the marble Prem Mandir to the world-famous ISKCON temple and the grove of Nidhivan; nearby Mathura, the birthplace of Krishna, is only about 14 km away.
Joyful bhakti. Alive with kirtan, aartis and the greeting Radhe Radhe, Vrindavan is a place of joyful, tender devotion and its Holi and Janmashtami are world-famous; it is the natural base for a pilgrimage of Braj.

What Barsana is
Barsana is the hill town of Radha Rani, in the west of Braj:
Radha's town. Barsana, about 45 to 50 km west of Vrindavan, is the home town of Radha Rani, the beloved of Krishna, the daughter of King Vrishbhanu; it is where Radha grew up and every corner is dear to her devotees.
The Radha Rani temple on the hill. The great Radha Rani temple, the Shriji temple, crowns the hill of Barsana, reached by a climb or a palki; there Radha, as Kishori Ji, is the presiding deity and her aarti is sung with the deepest love.
Lathmar Holi and the leelas. Barsana is world-famous for the Lathmar Holi, the stick Holi played with nearby Nandgaon, Krishna's town and is dearest of all on Radhashtami; it is a smaller, quieter hill town than Vrindavan, steeped in the love of Radha.

The key differences
In short, the two towns differ in a few clear ways:
Krishna's town or Radha's town. Vrindavan is Krishna's town, the main town of Braj; Barsana is Radha's town, her hill home in the west. So one is the town of the Lord and the other the town of his beloved, the two sides of one love.
The hub or a day trip. Vrindavan is the hub where pilgrims stay, with all the great temples and Mathura nearby; Barsana is a smaller hill town, usually visited as a day trip from Vrindavan, about an hour and a half away.
The size and the setting. Vrindavan is a large, busy temple town on the plains by the Yamuna; Barsana is a smaller, quieter hill town, crowned by its temple, in the western countryside of Braj, dear above all at Radhashtami and Lathmar Holi.
How they fit together
The truth is that Barsana and Vrindavan are not a choice but a pair, for the love of Radha and Krishna binds them. Most pilgrims base themselves in Vrindavan, the hub, with its temples and easy stay and from there make a day trip to Barsana, Radha's hill town, often with nearby Nandgaon, Krishna's town and Govardhan. So you take darshan of Krishna in his Vrindavan and of Radha in her Barsana and hold both halves of the love of Braj. To sing Radha's praises at Barsana and Krishna's at Vrindavan is to complete the pilgrimage; the marble prem mandir vrindavan, which honours Radha and Krishna together, carries the same love.
Which should you visit?
The honest answer is both; but here is how to think of it:
Base in Vrindavan. Vrindavan is the natural base, with its great temples, its stay and its nearness to Mathura; give it at least a day or two for Banke Bihari, Prem Mandir, ISKCON and the wider town and the Yamuna aarti.
Day-trip to Barsana. Barsana is a beautiful day trip from Vrindavan, for the darshan of Radha Rani on her hill, often with Nandgaon and Govardhan; it is dearest at Radhashtami and unforgettable at the Lathmar Holi.
See both for the whole of Braj. If you have only a day, Vrindavan is the essential base; but to truly know Braj and the love of Radha and Krishna, see both and if you can, the wider Braj of Nandgaon, Govardhan and Gokul too.
How to visit both, best time and tips
Barsana is easily visited from a base in Vrindavan:
Reaching Barsana. Barsana lies about 45 to 50 km west of Vrindavan and Mathura, about an hour and a half by road; most pilgrims visit on a day trip from Vrindavan, often with Nandgaon and Govardhan, by car with a Braj guide.
The climb. The Radha Rani temple crowns the hill of Barsana, reached by a climb of steps or a palki; wear comfortable shoes, carry water and allow time for the climb and the darshan, especially at festivals.
Best time. October to March is the most pleasant season; Barsana is dearest at Radhashtami, Radha's appearance day and unforgettable at the Lathmar Holi in the days before the main Holi, though very crowded, so plan well ahead for those dates.
