My advice for the drive is the same as I give everyone from the hills: start early, take one of the expressway routes rather than the slow all-highway one and plan a rest and meal near the Delhi ring. There are three main road routes from Rishikesh to Vrindavan; here they are side by side:

As in the other direction, the expressway is the sensible choice.
Which route should you choose?
For the quickest drive: the Yamuna Expressway route. In both directions the Yamuna Expressway route is the fastest, about 6 hours 43 minutes to Vrindavan and about 6 hours 29 minutes back to Rishikesh, on smooth, fast expressway for much of the way; it is the usual choice.
For the shortest distance to Vrindavan: the Eastern Peripheral Expressway. Coming to Vrindavan, the 361 km route via the Eastern Peripheral Expressway is the shortest, about 6 hours 48 minutes and helps you skirt the busiest parts of the Delhi region.
Best avoided: the all-highway route. The route via NH 344G alone, without the expressways, is slower in both directions, about 8 hours; choose it only if the expressways are closed or you have a particular reason.
Check live traffic on the day, since conditions near the Delhi region can decide which route is quickest; a driver who knows the roads will pick the best one.
How to travel between Rishikesh and Vrindavan
There are several ways to make the journey; the options at a glance:
By car or taxi (best). The most comfortable and flexible way, about 7 hours by one of the expressway routes, door to door, with rest stops of your choosing; ideal for families, elders and travellers with luggage and easily arranged as part of a Braj tour.
By train. There is no direct rail hop; you travel by train from the Haridwar or Rishikesh area to Mathura Junction, some services via Delhi and then cover the roughly 15 km from Mathura to Vrindavan by road. Check the current timetable and book ahead.
By bus. Buses run from Rishikesh and Haridwar towards Delhi and on to Mathura, from where you take a local ride to Vrindavan; it is the budget option, though slower and with changes.
By air. There is no direct flight for so short a hop; the nearest airport to Rishikesh is Dehradun (Jolly Grant) and to Vrindavan is Delhi, so road remains the practical way between the two.
